不同遗传背景小鼠输卵管移植效率的研究

Study on the efficiency of tubal transplantation in mice with different genetic backgrounds

摘要: 目的 选用不同遗传背景的小鼠,研究移植不同胚胎数量对产仔率的影响,旨在提高小鼠的移植 效 率。 方法 本研究主要采用输卵管移植技术将 2-细胞胚胎移植至假孕鼠体内,对妊娠母鼠的产仔数量进行统计分析。 结果 与封闭群相比,近交系的移植效率较低,平均产仔率为 37. 5%,封闭群则为 45%。 免疫缺陷动物对移植的胚 胎数量要求较高,每只小鼠单侧移植 11 ~ 15 枚细胞时的产仔率最高,为 63. 5%;不同类型小鼠的平均产仔率比较: C57BL / 6J(49. 8%) >KM(45. 8%) >ICR(44. 1%) >NOD-SCID( 35. 6%) >BAL B / c-nu( 29. 3%) 。 ICR、C57、NOD-SCID 三种小鼠移植胚胎数量在 11 ~ 15 枚时,产仔率最高;而 KM 小鼠移植胚胎数量在 16 ~ 30 枚时,产仔率最高;BALB / c-nu 小鼠移植胚胎数量≤10 枚时,产仔率最高。 结论 不同遗传背景的小鼠输卵管移植效率不同,相同遗传背景 小鼠移植产仔率随移植胚胎数量的不同而有所变化。

Abstract: Objective Effects of different numbers of embryos transferred in mice of different genetic backgrounds on the litter rate. Method The 2-cell embryos were transplanted into pseudopregnant mice by fallopian tube transfer technique. Then, Statistical analysis was performed on the litter size of pregnant animals. Result Compared with the closed colony, the transplant efficiency of the inbred strain animals is lower, the average litter rate is 37. 5%, and the closed colony is 45%. Immunodeficient animals have higher requirements for the number of embryos to be transplanted, and mice with 11-15 cells have the highest litter rate, 63. 5%; In this study, mice with different genetic backgrounds have different mean litter rate: C57BL / 6J ( 49. 8%) > KM ( 45. 8%) > ICR (44. 1%) > NOD-SCID (35. 6%) ; > BAL B / c -nu ( 29. 3%) . In this study, it was found that mice of different genetic backgrounds required different numbers of embryos to achieve higher litter rate. ICR, C57 and NOD-SCID mice need to transfer 11-15 embryos, KM mice need to transfer 16-30 embryos; BALB / c-nu mice need to transfer ≤10 embryos. Conclusion Tubal transplantation differs in mice with different genetic backgrounds. The litter rate of mice of the same genetic background varies with the number of transferred embryos.
